関東小桜一家のひとり娘・明美（みづなれい）は、やくざを嫌って東京の女子学園で寮生活を送っていたが、父が急死したという知らせを聞いて実家へ戻った。久しぶりに見る小桜一家はさびれ、子分もピン吉と捨松の2人きり。彼らは明美を姐さん、姐さんと呼んで、たてまつる。一方、小桜一家にかわって勢力を伸ばしてきた剛田組は、小桜一家の縄張りを乗っ取るため、数々のいやがらせを繰り返していた。明美を慕う妹分のチエ（若月まりあ）も剛田組のえじきに。怒りに燃える明美は2代目を襲名。セーラー服に日本刀を手に剛田組に殴りこむ……！
